چکیده

Purpose Abacavir use has been associated with an increased risk of ischemic cardiovascular events in several cohort studies, but the pathogenic mechanisms are still unknown. Recently Martinez et al. observed no differences in markers of inflammation, endothelial dysfunction, insulin resistance or hypercoagulability in HIV infected persons treated with either abacavir or tenofovir. While several studies have shown endothelial dysfunction in HIVinfected patients, only a few data are available on the involvement of platelets. Aim of our study was to evaluate markers of platelet activation and endothelial dysfunction in HIV infection comparing patients treated with abacavir or tenofovir.

Controversy about abacavir and tenofovir

In general, the risk of complications such as heart attack, stroke and poor blood circulation increases as people age. Among HIV-positive people, the risk of these complications is heightened compared to HIV-negative people because of the long-term effects of HIV infection. Chronic viral infections can cause inflammation, and ongoing inflammation can affect the health of many organ-systems, inc...

In vivo platelet activation and platelet hyperreactivity in abacavir-treated HIV-infected patients.

Abacavir (ABC) has been associated with ischaemic cardiovascular events in HIV-infected patients, but the pathogenic mechanisms are unknown. Aim of our study was to assess whether ABC induces in vivo platelet activation and ex vivo platelet hyper-reactivity. In a retrospective, case-control study, in vivo platelet activation markers were measured in 69 HIV-infected patients, before starting the...

Effect of abacavir on acute changes in biomarkers associated with cardiovascular dysfunction.

BACKGROUND This study examined the effect of abacavir on acute changes in biomarkers associated with cardiovascular dysfunction. METHODS Among the Study to Understand the Natural History of HIV/AIDS in the Era of Effective therapy (SUN) participants, we identified 25 individuals (cases) who were HLA-B5701-negative and who had ≥ 2 weeks without abacavir exposure at one visit and ≥ 2 weeks with...

Whole blood platelet aggregometry in HIV-infected patients on treatment with abacavir

Purpose: To assess platelet activity in HIV-patients with and without antiretroviral therapy (ART), analysing the influence of the presence or absence of ABC in the regimen. Methods: Observational, pilot study, including 30 asymptomatic HIV-patients: 20 on ART for at least 24 weeks and with undetectable HIV viral load—10 on ABC, 10 on tenofovir (TDF)— and 10 naïve patients, and a control group ...
